Application
The Kinetichrome heparin test kits are designed for the chromogenic measurement of the anti-factor IIa (thrombin) and/or anti-factor Xa activity in samples containing heparin, low molecular weight heparin and related molecules with thrombin and/or factor Xa inhibitor activity.
A distinctive feature of the Kinetichome product line is that these kits are designed and optimized for activity measurements with buffer as the reaction medium, rather than plasma. Thus, the Kinetichrome heparin test kits are ideally suited for research, industrial, and pharmacopeial test applications where buffer is the specified test medium.

The 8th edition of the European Pharmacopoeia, supplement 8.3, issued on 1st July 2014 and coming into effect from 1st January 2015 has new guidelines on chromogenic assays for anti-IIa and anti-Xa activities
Provision Kinetics has modified their Xa Heparin test kits in order to FULLY conform with the EP directives – accordingly, the XAE-100 and XAE-200 Heparin Test Kits are available now. The IIa Heparin Test Kits are already compatible the ruling.
Please note
The kits have 100 and 200 determinations when in optimal conditions on an automated haemostasis instrument, if you are using the manual test method(s) on a lab bench it is more realistic to expect about 50 determinations from a 100 determination kit and 100 from a 200 determination kit.

Summary and principle
Thrombin (Factor IIa) is the terminal enzyme in the classical coagulation cascade, where it plays roles in both the fibrin deposition and platelet activation processes of blood coagulation. Unfractionated Heparin (UFH) and Low Molecular Weight Heparin (LMW-H) inhibit blood coagulation via initial binding of these molecules to Antithrombin (AT), followed subsequently by the AT-heparin complex inhibiting Factor Ila. In buffer, this same “Anti-lla” activity of heparin can be foJlowed enzymatically by means of a chromogenic peptide substrate which, upon hydrolysis by residual factor IIa, provides a chromophore (405 nm) whose measure is inversely proportional to the initial amount of heparin present in the test sample. The paranitroanaline (pNA) chromophore generated by the residual factor lIa can be measured either kinetically or by an acid-stopped end-point method.
Schematically, the procedure is carried out as follows:
AT + heparin → (AT-heparin)
[AT-heparin) + IIa (known excess) → Ila (residual) + IIa-AT-heparin (inactive)
IIa (residual) + IIa Substrate (colorless) → pNA chromophore (405 nm) + peptide
